      目的 探讨同种异体脂肪组织来源干细胞(ASCs)联合地黄低聚糖(RGOs)对心肌梗死(MI)小型猪心肌凋亡的影响。方法 球囊封堵冠状动脉前降支制备小型猪MI模型,17头小型猪造模成功,采用随机数字表法分为4组:ASCs组(n=4),RGOs组(n=4),RGOs+ASCs组(n=4)及对照组(n=5)。造模前3天、造模后1月RGOs组及RGOs+ASCs组均饲以RGOs粗提物;造模7~10 d经冠状动脉移植ASCs,RGOs组及对照组均注入0.9%氯化钠注射溶液。移植后8周检测心肌梗死边缘区细胞凋亡(TUNEL法)及Bax、Bcl-2的表达。结果 与对照组比较,RGOs组及RGOs+ASCs组心肌细胞凋亡均显著降低(P<0.05,P<0.01),三组Bax的表达均明显降低(P<0.05),RGOs组及RGOs+ASCs组Bcl-2的表达均明显升高(P<0.01)。结论 ASCs联合RGOs抑制了梗死边缘区心肌细胞凋亡。

      Objective To investigate the effect of a combination of allogenic adipose tissue-derived stem cells (ASCs) and Rehmannia Glutinosa Oligosaccharides (RGOs) on the apoptosis of miniswine with myocardial infarction (MI).Methods MI was induced in miniswine hearts by occlusion of the left anterior descending artery for 90 minutes.The survived animals after MI were divided into four groups and received intracoronary ASCs (ASCs,n=4) 7 to 10 days later,intracoronary saline in combination with RGOs(RGOs,n=4) ((administration crude extract of RGOs from 3 days prior to MI to one month post MI),intracoronary ASCs in combination with RGOs (ASCs+RGOs,n=4),or remained untreated (control,n=5).The apoptosis of border zone and the expression of Bcl-2 and Bax were detected eight weeks after transplantation.Results The administration of RGOs significantly inhibited cardiomyocyte apoptosis of border zone in RGOs and RGOs+ASCs groups(P<0.05 and P<0.01).The expression of Bax in ASCs,RGOs and RGOs+ASCs groups was significantly decreased (P<0.05),and the expression of Bcl-2 in RGOs and RGOs+ASCs groups was significantly increased (P<0.01) compared to the control group.Conclusion The combination of ASCs and RGOs could reduce the apoptosis of peri-infarct myocardium.
