目的 探讨地黄低聚糖(RGOs)对过氧化氢-血清饥饿诱导的小型猪脂肪组织来源干细胞(ASCs)Bax、Bcl-2表达的影响。方法 过氧化氢(200μM)联合血清饥饿(H2O2/SD,6 h)建立小型猪ASCs凋亡模型。RGOs(0.01 g/L、0.1 g/L、1 g/L、10 g/L)预处理12 h并继续干预6 h。Western-blot法测定细胞Bax、Bcl-2的表达。结果 RGOs在一定浓度范围(0.1 g/L~10 g/L)可以减轻H2O2/SD引起的ASCs损伤,表现在Bax表达下调,Bcl-2表达上调。结论 地黄低聚糖对过氧化氢-血清饥饿诱导的脂肪组织来源干细胞的凋亡具有保护作用。 |

Objective To investigate the effect of Rehmannia glutinosa oligosaccharides (RGOs) on hydrogen peroxide-and serum deprivation-induced apoptosis in adipose tissue-derived stem cells.Methods An apoptosis model in miniswine adipose tissue-derived stem cells (ASCs) was established by hydrogen peroxide-and serum deprivation-induced (H2O2/SD,6h)in vitro.RGOs (0.01g/L,0.1g/L,1g/L,10g/L) pretreated for 12 hours and continued presence for 6 hours.Expression of Bax and Bcl-2 was investigated by using Western blot analysis.Results RGOs significantly attenuated hydrogen peroxide-and serum deprivation-induced ASCs apoptosis,showing a down-regulated Bax expression and an up-regulated Bcl-2 expression with the concentrations between 0.1g/L-10g/L.Conclution Rehmannia glutinosa oligosaccharides can protect adipose tissue-derived stem cells against hydrogen peroxide-and serum deprivation-induced apoptosis. |
