      目的 探究肠道益生菌补充对酒精、高糖、高脂致肝损伤大鼠肝功能和肠道菌群的影响。方法 高糖高脂饮食联合白酒灌胃处理制备酒精、高糖、高脂致肝损伤大鼠模型,并分为空白对照组、模型组、低剂量肠道益生菌组(20×108CFU /kg)和高剂量肠道益生菌组(40×108CFU /kg)。灌服白酒7周后,算大鼠脏体比、血清乙醇体积分数;检测血清中丙氨酸转氨酶(ALT)、门冬氨酸转氨酶(AST)、碱性磷酸酶(AKP)、三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平;使用酶联免疫吸附法(ELISA)检测肝组织中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、转化生长因子β1(TGF-β1)、二胺氧化酶(DAO)和D-乳酸(D-LA)水平含量;使用平板培养大鼠粪便中的乳酸杆菌、双歧杆菌、大肠杆菌、肠球菌,计算大鼠粪便中各种细菌的菌落数;使用Western Bolt测肝组织 NF-κB蛋白表达情况。结果 与空白对照组相比,模型组大鼠的脏体比显著升高,盲体比显著降低,体内乙醇含量显著升高,ALT、AST、AKP、TG、TC、LDL-C、TNF-α、IL-6、TGF-β1、DAO、D-LA含量显著升高,HDL-C含量显著降低,NF-κB蛋白表达量显著升高,且有统计学差异(P<0.01),低剂量组和高剂量组大鼠各项指标差异均无统计学意义(P>0.05);与模型组相比,低剂量组和高剂量组大鼠的脏体比显著降低,盲体比显著升高,体内乙醇含量显著降低,ALT、AST、AKP、TG、TC、LDL-C、TNF-α、IL-6、TGF-β1、DAO、D-LA含量显著降低,HDL-C含量显著升高,NF-κB蛋白表达量显著降低,且差异有统计学意义(P<0.01)。结论 补充肠道益生菌可以通过调节肠道益生菌的种类和数目、降低NF-κB蛋白的表达同时降低血清内的炎性因子达到保护酒精、高糖、高脂致肝损伤大鼠的肝功能和肠道菌群的效果。

      Objective To explore the effects of intestinal probiotic supplementation on liver function and intestinal flora in rats with liver injury induced by alcoholic,high glucose and high fat.Methods The rat model of liver injury induced by high glucose and high fat combined with liquor gavage was prepared,and divided into blank control group,modelgroup,low-dose intestinal probiotic group (20×108 CFU/kg) and high-doseintestinalprobiotic group (40×108 CFU/kg).After 7 weeks of liquor gavage,the organ-body ratio and serum ethanol volume fraction were measured.The alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase (AKP),triglyceride(TG),total cholesterol(TC),low-density lipoprotein sterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C) were detected.The tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),transforming growth factor-β1 (TGF-β1),diamine oxidase(DAO) and D-lactic acid(D-LA) of liver tissues were detected by enzyme linked immunosorbent assay (ELISA).The Lactobacillus,Bifidobacterium,Escherichia coli and Enterococcus in rat feces were cultured by the method of feces plate cultivation,and the quantity of various bacteria in rat feces was calculated.The expression of NF-κB protein in liver tissue was measured by Western Bolt.Results Compared to the blank control group,the organ-body ratio of model group was significantly increased,the blind body ratio was significantly decreased;the ethanol content in body,ALT,AST,AKP,TG,TC,LDL-C,TNF-α,IL-6,TGF-β1,DAO and D-LA of model group were increased significantly while the HDL-C was decreased significantly,and the NF-κB protein expression of model group was increased significantly(P<0.01),and there were no significant changes in all the indexes in low-dose group and high-dose group(P>0.05).Compared to the model group,the organ-body ratio of low-dose group and high-dose group were significantly decreased,the blind body ratio was significantly increased,and the ALT,AST,AKP,TG,TC,LDL-C,TNF-α,IL-6,TGF-β1,DAO and D-LA were significantly decreased while the HDL-C of low-dose group and high-dose group were significantly increased,and the NF-κB protein expression of low-dose group and high-dose group was significantly decreased (P<0.01).Conclusion Intestinal probiotic supplementation can protect the liver function and intestinal flora by regulating the types and quantity of intestinal probiotics,reducing the expression of NF-κB protein and inflammatory factors in serum in rats with liver injury induced by alcoholic high glucose and high fat.
