      目的 探讨OGP(10-14)及其类似物G48A对大鼠成骨细胞(OB)增殖、分化及cbfa1,IGF-1mRNA表达水平的影响,以期发现对骨质疏松的潜在治疗价值。方法 采用无血清培养条件,分别观察不同浓度OGP(10-14)及其类似物G48A对大鼠OB增殖和碱性磷酸酶(ALP)活性的影响;逆转录-聚合酶链反应检测OGP(10-14)及其类似物G48A对无血清培养大鼠三代OB cbfa1,IGF-1 mRNA表达的调节。结果 透射电镜下观察,OGP(10-14),G48A和PTH组OB呈成骨相,而CON组OB呈退化状态;OGP(10-14)和G48A对大鼠OB的增殖和ALP活性调节存在浓度-效应关系,OGP(10-14)和G48A组大鼠OB cbfa1,IGF-1 mRNA的相对表达水平较CON组显著上升(P<0.05)。结论 OGP(10-14)及其类似物G48A在体外具有促进大鼠OB增殖与分化的作用,OGP(10-14)和G48A干预上调大鼠OB cbfa1,IGF-1mRNA的表达。OGP(10-14)和G48A可能在调节成骨细胞功能方面发挥了重要作用。

      Objective To explore the effects of OGP10-14 and its analogue G48A on proliferation of OB and the relative expression level of cbfa1,IGF-1 mRNA in osteoblasts,so as to find out the potential therapeutic value for osteoporosis.Methods Primary osteoblastic cells were harvested derived from calvarial bone of new born SD rats.Subconfluent rat OB of 3 passage were perfomed quiescent by placing them in serum-free α-MEM culture medium for 24 hours and then were divided into the control and experimental groups cultured in the serum-free medium containing OGP10-14 and G48A in various concentrations.The proliferation of osteoblasts and activity of alkaline phosphatase of culture supernatant were determined.The relative levels of cbfa1 and IGF-1 mRNA expression were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR).Results The observation of transmission electronic microscope showed that the OB in OGP(10-14) and G48A groups were developed into osteogenic phase and the OB in the control group developed into degenerative phase.In vitro,OGP(10-14)and G48A can induce the concentration-effect relationship on OB proliferation and activity of alkaline phosphatase.Comparing with control group,the mRNA expression of cbfa1 and IGF-1 were significant increased in intervention groups(P<0.05).Conclusion OGP(10-14) and G48A can enhance proliferation and alkaline phosphatase activity of OB cultured in vitro.The expression of cbfa1 and IGF-1 were up-regulated by OGP(10-14) and G48A,therefore,OGP(10-14) and G48A may play an important role in regulating osteogenic function of OB.
