      阿尔茨海默病(AD)是一种以进行性中枢神经系统退行性改变为典型特征的疾病,致病原因及其病理机制十分复杂,临床上主要表现为出现认知功能障碍和学习记忆能力减退。小檗碱(BBR)是黄连的主要生物碱成分,临床应用广泛,越来越多的研究表明其对AD的防治具有一定作用。本文概述BBR的化学结构与其药代动力学特性,重点探讨其多靶点防治AD的分子机制及递送载体改良策略,深入剖析BBR通过调控肠脑轴干预AD的作用机制;回顾近5年来国内外关于BBR 防治AD的实验研究,重点分析BBR在“多靶点调控-递送技术革新-肠脑轴整合”三维度的独特价值,为BBR防治AD提供参考依据。

      Alzheimer′s disease (AD) is a progressive neurodegenerative disorder characterized by complex etiological factors and pathological mechanisms,primarily manifesting as cognitive dysfunction and impaired learning and memory capabilities in clinical settings.Berberine (BBR),the principal alkaloid constituent of Coptis chinensis,has been extensively utilized in clinical practice.Recent studies have increasingly demonstrated its potential therapeutic efficacy in the prevention and management of AD.This paper provides a comprehensive overview of the unique isoquinoline chemical structure of BBR and its pharmacokinetic properties,with a particular focus on elucidating the molecular mechanisms underlying its multi-target approach in preventing and treating the core pathologies of AD.It also examines the strategies for optimizing drug delivery systems and conducts an in-depth analysis of the mechanisms by which BBR intervenes in AD through the modulation of the gut-brain axis.A comprehensive review of the experimental research mechanisms on the prevention and treatment of AD with BBR over the past five years has been conducted,with a particular focus on analyzing BBR′s unique value across three dimensions:multi-target regulation,innovative delivery technologies,and gut-brain axis integration.This review provides a reference for the application of BBR in the prevention and treatment of AD.
