      目的 探讨大蒜素对大鼠肾缺血再灌注损伤(IRI)过程中的保护作用及机制。方法 将40只Wistar大鼠随机分为4组:假手术组(A组)、假手术+大蒜素组(B组)、肾脏IRI组(C组)、肾脏IRI+大蒜素组(D组)。HE染色观察肾脏组织病理变化;测定丙二醛(MDA)、超氧化物歧化酶(SOD)含量;采用原位缺口末端标记检测肾小管上皮细胞凋亡;免疫组织化学染色测定Bax和caspase-3的表达;Western Blot法检测Bcl-2和Cyt-C蛋白表达水平;qRT-PCR法检测肾脏组织中的Bax、Bcl-2、Cyt-C和caspase-3 mRNA表达水平。结果 与A组和B组相比,C组可见肾小管扩张和充血,肾小管上皮细胞肿胀和坏死等病理变化;MDA和凋亡水平显著升高,SOD活性显著降低;Bax、Cyt-C和caspase-3的表达水平显著增加,而Bcl-2的表达水平明显减少。与C组相比,D组中显示肾脏病理损伤明显改善;MDA和凋亡水平显著降低,SOD活性显著升高;Bax,Cyt-C和caspase-3 的表达水平明显减少,而Bcl-2的表达水平明显增加,差异具有统计学意义。结论 大蒜素可减轻氧化应激水平,发挥抗凋亡作用,从而在肾脏IRI过程中发挥保护作用。

      Objective To investigate the protective effect and mechanism of allicin on ischemia reperfusion injury (IRI) in rats.Methods Forty Wistar rats were randomly divided into 4 groups:sham-operated group (group A),sham-operated + Allicin group (group B),kidney IRI group (group C) and kidney IRI group + Allicin group (group D).HE staining was used to observe the pathological changes of kidney tissues;the contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured;the apoptosis of renal tubular epithelial cell was detected by DUTP Nick end labeling;the expression levels of Bax and caspase-3 were detected by immunohistochemistry staining.The protein expression levels of Bcl-2 and Cyt-C were measured by Western-Blot method.The Bax,Bc1-2,Cyt-C and caspase-3 mRNA expression levels in kidney tissues were detected by qRT-PCR method.Results Compared with group A and group B,group C showed pathological changes such as renal tubular expansion and hyperemia,renal tubular epithelial cell swelling and necrosis.The levels of MDA and apoptosis were significantly increased and the activity of SOD was obviously reduced.The expression levels of Bax,Cyt-C and caspase-3 were obviously increased,while the expression level of Bcl-2 was significantly decreased.Compared with group C,group D showed marked improvement in renal pathological injury.The levels of MDA and apoptosis were significantly decreased and the activity of SOD was obviously increased.The expression levels of Bax,Cyt-C and caspase-3 were obviously decreased,while the expression level of Bcl-2 was significantly increased.These differences were statistically significant.Conclusion Allicin can reduce the level of oxidative stress and play an anti-apoptotic effect,which can play a protective effect in the process of kidney IRI.
